My wife has purchased a flat from her brother in the year 2009 in Delhi by executing regd POA and will and motorised Sale deed now she wants to execute a regd sale deed.Can she do that.Also pl let her know the amount to be spent on stamp duty etc

Any transaction involving transfer of property needs to registered. Stamp duty varies from State to State.

POA holder can not register / sell the property to himself.

stamp duty differs from state to state, confirm from local lawyer.
