Recently one client approached me with one non judicial stamp paper worth of Rs.50/- of 1959 i.e., simple sale deed, but I found the signatures are some what difference. So please advise me that whether the Rs.50/- Non judicial stamp paper is available in the year 1959 or not. I want to confirm Whether it is genuine or not. Thats all.

get certify copy of sale deed from sub-registrar office and match the same.
or enquire locally at sub-registrar/adm(F&R)/Collector office.

very difficult to answer whether in 1959 stamp paper of rs 50 was available . try sub registrar office . you may succeed .

This is not a quiz.
But I guess stamp paper for Rs. 50/- was available at that time.
I vaguely remember seeing one old sale deed on Rs. 100/- stamp paper (Total Value Rs. 102/-) and was wondering. Though I am not sure it was of which year.
Even now stamp papers are available in Rs. 10000/- and above denomination for which even currency notes are not available.
