I want to prepare leave and lease agreement of the office premises in mumbai month rent of Rs. 66000/-

Can I prepare it in 100/- Rs. Stamp paper

No. This should be registered paying full stamp paper.

No, depending upon the sum of rent for twelve month period and security deposit amount, stamp duty @ 6,000/- or 10,000/- for each term of 12 month or part thereof shall be payable in your case. The agreement needs to be registered also.

You being the landlord in your safer interest of your property it is always in your good interest to get register the leave and license agreement.

whether you want to enter in leave and license agreement or lease agreement pls.clear as definations in law for both are different in case it is a lease then in my opinion you should follow the calculation enquiring at subregistrar office in your area.
