I've been told that, with reference to initiating a Summary Suit for recovery in Mumbai, if the Defendant is doing business in another state, I first need to seek leave of the court to file a Summary Suit. If yes, please direct me to the applicable Order and/or Rule under CPC.

No. you need not seek leave to file summary suit. you need to file summary suit before Court under whose jurisdiction your clients Registered Office or Residence is situated.
