Is there any rule to give preference to internal candidate in promotion ?

There will be an interview for promotion to the Internal Candidates and those who successfully complete the interview may select for promotion.

To the best if my knowledge, promotion of existing employee to higher position and recruiting directly to that position depends upon internal rules of the company/organization.

Yes, every department has "out of turn quota" for promotion of the candidates such as olympic winners etc.
